Recent Publications: Assessing Antibody Pharmacokinetics in Mice with In Vivo Imaging
2011-05-31
inviCRO announces the publication of a recent article in the Journal of Pharmacology and Experimental Therapeutics authored by Dr. Hoppin, Dr. Orcutt, Dr. Hesterman, and Dr. Lackas at inviCRO along with collaborators at the University of Massachusetts Medical School and Millennium Pharmaceuticals. The article, Assessing Antibody Pharmacokinetics in Mice with In Vivo Imaging, was selected for the cover story in the May 2011 edition of JPET. The article reviews basic principles of radiolabeling and imaging methods relevant to antibodies with a focus on practical considerations of study design and execution. The article includes detailed tables with information on imaging sensitivity, resolution, radiochemical properties, and cost of common SPECT and PET isotopes. Images and analysis from a case study are presented. Image data were processed to assess the biodistribution of 111In-labeled specific and non-specific antibodies in xenograft tumors and other regions of interest at select times out to 96 hours post-injection. The figure displayed on the left illustrates the powerful qualitative and quantitative advancements that have been made in the field of small-animal molecular imaging. A, maximum intensity projections of SPECT (color) and CT (gray-scale) of two xenograft mice injected with a 111In-labeled specific antibody (top) or 111In-labeled nonspecific antibody (bottom). B, surface plots displaying radial tumor profiles of activity distribution as a function of time for the specific antibody (left), nonspecific antibody (middle), and merge (right).
